Different hormone stimulus during pro-estrous after synchronization of ovulation programs on uterine receptivity for gestation of suckled zebu females.

Abstract

This project aims to evaluate different hormonal stimulus during the pro-oestrus to manipulate the occurrence of estrus and pregnancy on uterine receptivity in suckled zebu. For this, two experiments are being proposed. In Experiment 1, will evaluate the effect of proestrous supplementation with estradiol cypionate (ECP) or equine chorionic gonadotropin (eCG) on the global gene expression in uterine tissue at subsequent diestrous following synchronization of ovulation. A total of 60 zebu females will receive a synchronization of ovulation protocol for timed artificial insemination (TAI). At progesterone (P4) device withdrawal, females will be assigned into one of four treatments. Females of the control group will not receive any additional treatment, while females of ECP or eCG groups, will be treated with 1mg of ECP or 400 IU of eCG at moment of P4 device removal, respectively. Females from ECP+eCG group will be treated with both treatments. All females will be treated with GnRH 48 hours after P4 device removal. Seven days after GnRH treatment, females will undergo uterine biopsy for evaluate the genes expressed in the endometrial tissue. In experiment 2, females (n=200) will receive the same treatments described in Experiment 1, however, in this experiment will be quantified the amount transcript genes differently expressed by real time PCR. Furthermore, the expression of such transcript genes will be retrospectively associated with the occurrence of estrus and the pregnancy success. Therefore, could be suggested an existence of an optimum uterine receptivity environment for gestation and its receptivity could be manipulate pharmacologically.
