Abstract

Introduction: It is inevitable that an increasing number of patients with renal or not submitted at some point in life surgeries. In 2004 there was a pattern to acute alteration of renal function through the RIFLE criteria. The criteria relate to glomerular filtration rate (serum creatinine) and urine output with the clinical outcome, therefore with the severity of renal injury. However, the serum creatinine used for the diagnosis of the lesion is not the most reliable biomarker for analysis during the acute changes in renal function.In search of more sensitive biomarkers for early detection of acute renal dysfunction, the associated lipocalin neutrophil gelatinase (NGAL), in both plasma and urine, appears as an independent predictor. NGAL protein expression is induced by epithelial injury and in animal models after ischemic or nephrotoxic renal injury was confirmed early and marked his elevation 8.9. Through methods such as Western blot and ELISA (enzyme-linked immunosorbent assay) were labeled plasma levels and urinary NGAL with an increase of ten times or more during the period between 2-6 hours post-cardiac surgery in patients who subsequently developed acute kidney injury. Objectives: To evaluate the protective effects of dextrocetamina on renal function, using an experimental model of ischemia and reperfusion in rats by means of serum creatinine, NGAL and interleukins. Methods: After submission of this research project to the Ethics Committee on Animal Experiments of the Faculty of Medicine of Botucatu, will be included in the study, 40 Wistar rats, weighing more than 250g, provided by the Central Animal Facility of UNESP Botucatu campus.Will be divided into 04 groups: Group S or Group Sham (n = 10): SD Group or Group with Dextrocetamina Sham (n = 10): Group C or Group Control (n = 10): Group E or Group Study (n = 10), which will be anesthetized with the inhalational anesthetic isoflurane as described in "Experimental Sequence" of the project and will also be performed laparotomy and right nephrectomy in four groups of animals. Also we will study the attributes, PS, DP, MAP, HR, rectal temperature from catheterization of the right carotid artery and at times presented as highlighted in the experimental sequence. The plasma NGAL as well as creatinine and interleukins are evaluated at M1, M2, m3, m4. The attributes studied in each animal of the four study groups the following attributes: Systolic Pressure (SBP), diastolic (SD) and mean (MAP), heart rate (HR), rectal temperature, plasma NGAL Dosage, Dosage plasma interleukins and measurement of plasma creatinine.Statistical analysis of data obtained will be used to profile analysis. The histological results will be analyzed by the non-parametric Wilcoxon test for paired samples and the nonparametric Mann-Whitney test for independent samples.
