Pregnancy rate, inflammatory response and uterine hemodynamic after artificial insemination with cryopreserved semen evaluated by fluorescent probes

The aim of this study was to verify the pregnancy rate and the inflammatory response in mares, considering the uterine hemodynamic and endometrial cytology after artificial insemination (AI) with frozen semen of high and low quality, determined by the plasma and acrosomal membranes integrity , and mitochondrial function of the spermatozoa using fluorescent probes. Semen samples of one stallion would be evaluated by the association of the fluorescent probes: PI-H342, FITC-PSA and JC-1 and would be separated according to the percentage of cells with plasma membrane integrity, acrosomal integrity, and with mitochondrial function (PIAIC). Sixteen previously selected mares would be used, discarding those with predisposition for endometrites, considering reproductive performance, and gynecologic exam. The mares would be separated in two groups for a first reproductive cycle being: Group C (control): not inseminated mares, with mimic of AI procedure (n=8), Group D: intrauterine infusion of extender (n=8). The same mares would be separated for a second reproductive cycle, being: Group A: mares inseminated with high quality semen (n=8), and Group B: mares inseminated with low quality semen (n=8). Follicular development would be monitored daily, in the presence of a 35mm follicle, human chorionic gonadotrophine (hCG) would be administrated to induce ovulation. The uterine evaluation would be done by color Doppler ultrasonography in seven moments: immediately before hCG administration (T-30), immediately before insemination (T0), 2 (T2), 6 (T6), 12 (T12), 24 (T24) and 48 hours (T48) after insemination. The uterine cytology would be done at 12 hours after AI. Pregnancy diagnosis is going to be done 14 days after AI by B-mode ultrasonography. The results of fertility would be evaluated by X2 test and the inflammatory response resulting from the quality of semen would be performed by the SAS program (SAS,2010) with the regression analysis.
