Metabolomic and proteomic profile and cell proliferation and apoptosis in Walker tumour-bearing rats under pregnancy hormones and leucine-rich diet

Pregnancy is characterized by many maternal physiological adaptations related to hormonal variations levels, especially oestrogen, progesterone, hCG and prolactin, which are essential to support foetal growth. Pregnancy hormones influence and modulate the breast cancer incidence and growth. However, there are few source and controversial data about the influence of these hormones on tumour growth. For this reason, this study aim to investigate the influence of pregnancy hormones - oestrogen, progesterone, hCG and prolactin - and also the influence of nutritional supplementation with leucine on Walker 256 tumour growth in vitro and in vivo. In vivo studies will be conducted in Wistar rats, which will be distributed in eight different groups: control (C), Walker tumour-bearing (W), pregnant (P) and tumour-bearing pregnant rats (PW); additional four groups subjected to leucine-rich diet; leucine (L), leucine tumour-bearing (LW), leucine pregnant rat (LP), leucine tumour-bearing pregnant rat (LPW). The inoculation of Walker 256 tumour occurs in the second day of pregnancy, and after 19 days of experiment, the animals will be sacrificed to analyse the metabolomic profile, serum hormones and cytokines content and tumour tissue will be analysed the profiles metabolomic, proteomic expression of proteins related to signalling processes, cell proliferation and apoptosis. In vitro experiments, Walker 256 cells culture will be treated with pregnant hormones and will be accessed and evaluated these cells proliferation and cytotoxicity under pregnant hormones, and also the effects on cell signalling proteins and apoptosis.
