Arterial hypertension is a complex syndrome mediated by genetic and environmental factors. Because several interacting mechanisms are involved in the maintenance of hypertension the control of high blood pressure (BP) is still a challenge. The sympathoexcitation and the renin angiotensin (Ang) system activation are main targets in the treatment of hypertension, thus, our aim is to understand the interaction between these systems in an experimental model of hypertension. Our previous studies showed that after 10 days of total renal denervation the BP is reduced in an Ang II-dependent model of hypertension, the 2 kidney-1 clip (2K-1C) model in Wistar rats. Hence, the main objective of this proposal is to study the renal afferent and efferent activity, separately, by selective renal denervation in an animal species that allow chronic recordings of BP and two sympathetic drives, simutaneously, to the kidney and to the heart. Selective afferent and efferent denervation will be performed in intracerebroventricular (icv) Ang II-infused sheep and BP, renal sympathetic nerve activity (rSNA) and cardiac sympathetic nerve activity (cSNA) will be measured for 14 days in conscious sheep. Ang II receptors type 1 (AT1) and type 2 (AT2) will be quantified in the kidneys and Ang II, plasma renin activity, Na+, K+, proteinuria, urea and creatinin clearance will be measured during icv Ang II infusion and selective renal denervation. Therefore, the present proposal intends to understand the mechanisms by which the Ang II and sympathetic activation interact and contribute to the maintenance of hypertension. (AU)
