The proteomic is the expression of the organism proteome study, or final genes expression result, and can be used evaluate the probiotics effects on fish immunity. The DGGE is a technique that can allow tracing of bacteria in fish intestines, and modifications of the intestinal flora caused by probiotic supplementation. Seven hundred and twenty Nile tilapia (15g) will be stocked in 16 tanks (800 L). The experimental design is randomized with three treatments (continuous probiotic, probiotic and probiotic-07 pulse pulse-14) + control and four replicates. The fish will be fed continuously with a diet containing probiotics (1.0 g kg-1 of Enterococcus faecium - 109 CFU g-1), feed on pulse-07 (seven days of feeding with probiotic and seven-day diet without probiotic) and pulse-14 (14 days with food + probiotic and 14 days without probiotic, until at the end of experiment). The livers of eight fish per replicate will be removed, transported to the University of Malaga/Spain and submitted to proteomic analysis. The intestines of the eight fishes will be removed and extracted mucus, freeze-dried and transported to the UMA to perform DGGE analysis. The project aim to evaluate the immune status using proteomic study of liver and intestinal microbial changes of Nile tilapia fed with probiotic Enterococcus faecium, develop and adapt the techniques of proteomics and DGGE for Nile tilapia, with researchers of the Department of Microbiology, University of Malaga, incorporating new techniques for evaluation of probiotics in scientific research on fish in Brazil.
