Mangroves are considered an unique ecosystem, due to its particular combination of environmental conditions, influenced by its location at the interface between land and ocean. The study of this ecosystem becomes urgent since mangroves are disappearing worldwide, and its diversity of microbial groups are still poorly known. Within this theme, this project aims to describe the possible functionality of bacteria belonging to the phylum Planctomycetes in the mangroves. Such organisms are still poorly studied, difficult cultivation and slow growth and / or dependent on the interaction with other micro-organisms. Within this phylum are found microorganisms belonging to genera described as capable of anaerobic ammonium oxidation (anammox), which is an important transformation of nitrogen in environments with low oxygen availability, such as mangroves. It is estimated that about 50% of the N2 is formed from marine process anammox. In this context, the metagenomics allows the exploration of potential biological independently of cultivation, by cloning of large DNA fragments obtained from environmental samples. In this project, the focus will be the detection and characterization of sequences of these organisms in clone libraries that make up metagenomics ever built, the sediment samples from the mangroves of Sao Paulo. It is hoped that this methodology, using PCR to detect specific sequences related to Planctomycetes group of clones metagenomics mangroves, and describe the genes found in such inserts in an attempt to turn the phylogeny of this group the different functions that it can have on maintenance and functionality of this environment.
