Smoking is the major risk factor for the development of chronic obstructive pulmonary disease (COPD), which is associated with high morbidity and mortality and use of health resources. In recent years studies have shown that regular aerobic training in smokers reduce the decline in lung function and the risk of developing COPD throughout the years. Recent experimental study developed by our group showed that the mechanisms related to the protection of physical exercise in the development of emphysema involve anti-inflammatory mediators and antioxidants. However, the intracellular mediators of these exercise effects on the development of pulmonary emphysema have not been investigated. Thus, the present study aims to evaluate the temporal effects of Nfr2-Keap1 proteins signaling on the lungs and peripheral skeletal muscle as possible modulators of the anti-inflammatory and antioxidants effects of aerobic training in an experimental model of pulmonary emphysema. The smoke group will be exposed to cigarette smoke for 30 minutes / day and the animals from exercise group will undergo physical training of moderate intensity 60 minutes/day. Both protocols will be carried out: 5 days/week for 4, 8 and 12 weeks. The parameters evaluated are: lung mechanics, number of total and differentials cells in bronchoalveolar lavage fluid, alveolar diameter, remodeling of the airways and lung parenchyma, determining levels of TNF-a, IL-1beta, IL-6, IL-10 and IL-1ra; levels of superoxide anion and hydrogen peroxide (oxidizing) and the levels of antioxidant enzymes (superoxide dismutase, glutathione peroxidase and catalase) in lung and muscle tissue.
