The present proposal aims to develop a miniaturized fiberoptic sensing system combining a luminescent sensor (LS) with a mid-infrared hollow wave guide (MIR HWG) sensor for the simultaneous determination of biomarkers such as total CO2 and O2 and volatile organic compounds (VOCs) (e.g. NO, acetone and the ratio 13C/12C) in breath samples. The IR sensor will determine molecular constituents and the fluorescence sensors will provide vital information on total O2 and CO2. The sensor platform will be develop and adapt toward real-time in situ and non-invasive continuous measurements in respirator systems in intensive care unit. For data classification and also analyte quantification in real time tracing biomarkers panels, multivariate data analysis will be employed. Initially breath samples analysis of mice and pigs at intensive care unit, available at Clinics for Anesthetics at Ulm Univ. within the requirements of the Ethics Committee, will be accomplished offline and on-line in order to evaluate the miniaturized fiberoptic sensing system performance. For validation purposes, conventional GC-MS breath analysis will be carried out and figures of merit carefully evaluated. As the final goal of this study is to establish an IR-HWG-Fluorescence tool for online monitoring of human patients in intensive care environments, measurements in humans exhaled breath (EB) will be carried out in collaboration with Lung Cancer Center at the Robert-Bosch Hospital Stuttgart. The proposed research is considered pioneer in breath diagnostics which for the first time aims at combining two orthogonal fiberoptic sensing concepts into a single diagnostic platform, and represents a fundamental step toward the increased adoption of optical chem/bio sensing techniques capable of on-line monitoring in clinical diagnostics. Also, innovation resulting from this project will be the first combination of IR and fluorescence sensing schemes into a single device enabling advanced breath diagnostics, opening up new opportunities for non-invasive continuous monitoring of breath analysis.
