Abstract

In previous study we showed cachexia establishment in solid Ehrlich carcinoma bearing (SET) mice from the tenth day of tumor inoculation which is associated with a severe decrease in insulin secretory capacity. This profile led us to evaluate the inflammatory condition of pancreatic islets in SET mice. The results showed that tumor bearing mice had increased protein expression of pro-inflammatory cytokines and in toll like receptors 3 and 4. High levels of free fatty acids on plasma, as shown in our tumor model, can increase TLR4 content in human monocytes, leading to an augmentation cytokine production and inflammatory condition. However, there are no studies that seek to correlate the inflammation of the endocrine pancreas with fatty acid content in islet. It is known that inflammation and free fatty acids can act together in the development of cachetic syndrome. The aim of this study will be evaluate the lipid content in the islets of Langerhans of solid Ehrlich carcinoma bearing mice searching for mechanisms of inflammation establishment in the endocrine pancreas, which seems to be key factors in the establishment of cachetic syndrome in tumoral models. For that, male adult Swiss mice (45 days old) will be inoculate with solid Ehrlich tumor (SET) and will be sacrificed 14 after tumor implantation whilst control group will receive saline alone (CTL). Pancreas will be fixed in bowan and paraffin embedded to histological analysis by immunohistochemistry. The results will be expressed in mean±SEM. Significance of the differences will be evaluated of t-test . The significance level will be P<0.05. (AU)
