Both, information obtained by our lab on the role P5CDH in Trypanosoma cruzi metabolism and biology, and available information in the literature support the role of this enzyme in several physiological processes in the African trypanosome (T. brucei) and its intermediate host Glossina morsitans (tsetse fly). This proposal intends to assess the phenotype of conditioned mutant null cells for Tbp5cdh gene (by using the RNAi technique) with respect to its ability to establish the infection in the controlled-infection model T. brucei - G. morsitans (in vivo). This model has been well standardized in the Liverpool School of Tropical Medicine (LSTM) and is maintained and used by Prof. Álvaro Acosta-Serrano, who participates of this proposal. In parallel, the analysis of possible interactions between TbP5CDH and other proteins will be performed by using the TAP-tagging method. To reach the proposed objectives, mainly the involvement of TbP5CDH in the T. brucei - insect host interaction, some tools already developed along my PhD will be used (such as: substrate, antibodies, enzymatic inhibitor, oligonucleotides, etc.). The information to be obtained will constitute a relevant contribution to understand the logics underlying the metabolism of different trypanosomatids of sanitary relevance, and will be also included in my PhD thesis.
