Abstract

Generation of induced pluripotent stem cells (iPS) from somatic cells has shown that adult mammalian cells can be reprogramed to a pluripotent condition through the implantation of some embryonic transcription factors. This discovery has raised fundamental matters about the mechanisms that, through this transcription factors, influenced cells epigenetically and its differentiation potential, during reprograming and normal development. Lipids and lipoprotein components affect some aspects of cell behavior during its differentiation and survival. This components act mainly through the activation of membrane receptors, responsible for activating cytoplasmic regulatory factors and stimulating or blocking specific signals of transduction pathways. Specific chromatin modulations and the activity of transcriptional factors are directly influenced by these signals (Chawla et al., 2001; Stulnig et al., 2003; Rawson et al., 2003). Hence, specific study about cellular and molecular mechanisms involved on reprograming became essential for major understanding of this process, thus, are necessary for better characterization and efficiency. Therefore, this study's proposal is the characterization of iPS cells from human mesenchymal cells and the investigation of the genetic induction at the lipidic level (lipidic profile). To do so, mRNA frequency of genes known as essential to pluripotency maintenance will be analyzed, as well as the definition of the lipidic profile during cells pluripotency induction process and differentiation process, after colonies were obtained.
