Thiazolidinedione (TZDs) are drugs widely used for the treatment of patients with type 2 diabetes. Despite its hypoglycemic activity, the TZDs also have pleiotropic effects on the vasculature. Indeed, TZDs, including rosiglitazone and pioglitazone, can inhibit endothelial cell migration and angiogenesis, a process associated with diabetic complications. However, despite the beneficial effects reported with treatment with these compounds, adverse effects are also reported. Importantly, weight gain, increased risk of fractures as well as an increased risk of heart failure are associated with TZD therapy. Therefore, the development of new TZDs with similar beneficial effects and fewer adverse effects is needed. The present study will investigate the effects of two new arylidene-TZDs, called GQ2 and GQ11 in endothelial cells and in cardiomyocytes derived from stem cells. To achieve the goals, models that mimic the different stages of the angiogenic process will be employed. The analysis of the new TZDs effects on the expression of angiogenic genes will also be performed. In addition, the cardiotoxic effect of the new TZDs will be explored in cardiomyocytes derived from stem cells.(AU)
