This study aims to validate different models of nociceptive stimuli in horses, establish a comparison between the different types of stimuli as well as determine a valid and reliable method to evaluate the effectiveness of analgesic drugs in this species. Eight adult horses will undergo three intravenous treatments: control (saline), MD (methadone 0.2 mg/kg + detomidine 10 mcg/kg) and MA (methadone 0.2 mg/kg + acepromazine 0.05 mg/kg) administered randomly and with a minimum interval of one week between each treatment. The antinociceptive activity will be evaluated using the voltage, temperature and mechanical pressure to the withdrawal reflex of the limb after electrical, thermical and mechanical stimulation. Electrical stimulation Will be performed with an electrical stimulator attached to the coronary band of the thoracic limb of the animal, starting with 1V and increasing until a nociceptive response is observed. The mechanical and thermal stimuli will be assessed with wireless devices, in which mechanical and thermal values will be recorded automatically on a digital display. To evaluate the mechanical stimulus, the probe will be fixed on the dorsal region of the third metatarsal bone of the animal and the pins will be pressed against the skin by means of pneumatic devices. For the evaluation of the thermal nociception, the probe will increase the temperature until the animal responds. (AU)
