Abstract

Currently some systems have been developed for intraocular sustained release througH because there are several barriers to achieve the therapeutic concentration and excretion is usually very fast, so they have to repeat many times the administration, such as direct injection. Another system of extended release of drugs is the micro / nanoparticles with a specific polymer obtained by the technique of "spray drying" to keep the concentration of drug inside the eye for a longer period. There are two main types of eye tumors, retinoblastoma usually found in children and choroid melanoma in adults, and both, if left untreated can lead to blindness. Methotrexate (MTX) is an anti-neoplastic analogue of folic acid and dihydrofolate reductase inhibitor. The action of MTX occurs in cells in exponential growth phase. It also has anti-inflammatory and immunosuppressive effect in the case of rheumatoid arthritis and psoriasis. It aqueous solubility is very limited, thus hindering their therapeutic efficiency. This work aims to develop micro / nanoparticles of biodegradable polylactic acid-co-glycolic acid (PLGA) methotrexate satisfaction and evaluate their potential in vitro and drug delivery system for intraocular administration.The micro / nanoparticles are obtained by spray drying technique in "spray dryer". The drug-polymer interactions will be studied using absorption spectroscopy in the infrared (IR), differential scanning calorimetry and thermogravimetry derivative. Is it calculated the amount of drug incorporated, as well as validation of methods of quantification.
