Abstract

The local anesthetic toxicity has been shown in many cell types and may be associated to the production of proinflammatory cytokines. Articaine has been implicated in many cases of paresthesia after inferior alveolar nerve block; this occurrence seems to be concentration-dependent. The association to carriers (objective of the project "temático" associated), such as liposomes, has been shown to reduce systemic toxicity, to increase the plasmatic half life "in vivo" and to increase anesthesia duration in some models. Therefore, the aim of this study is to analyze the effects of articaine on the modulation of IL6 and IL1± cytokines, on caspase 3 and on the celular viability in human gengival fibroblats. The cells will be exposed to 2%, 3% and 4% plain articaine, 2%, 3% and 4% articaine encapsulated into unilamelar liposomes and controls (saline, liposomal suspension and without treatment). IL6 and IL1± cytokines (Elisa immunoassay), caspase 3 (fluorimetric assay) and cellular viability (XTT reduction - spectrophotometry) will be evaluated at 10min and 4h after exposure to the treatments. Statistical analysis will be performed with BioEstat 5.0. Data will be submitted to ANOVA (Tukey test) or Kruskal-Wallis (Dunn test) depending on normality of distribution and homocedasticity of variances. Significance level will be set at 5%.
