Insects are responsible for the transmission of infectious diseases that affect humans and other animals, including dengue, malaria, leishmaniasis and yellow fever among others. To overcome the barriers imposed by vertebrate hosts and be able to feed, haematophagous mosquitoes possess salivary components capable of regulating hemostasis, inhibit the inflammation and modulate the host immunity. Since the 80's several studies have demonstrated the anticoagulant and vasodilator properties of components present in the salivary gland of the mosquito vector of dengue, Aedes aegypti. The study of proteins and mRNAs present in its salivary glands (sialome) has exponentially increased the description of the molecules that compose the saliva cocktail. However, the characterization of the biological activities of these molecules in the host still needs further studies. By using bioinformatics tools, we have observed the presence of a serpin family, inhibitors of serine protease, in Ae. aegypti sialome. In this project we intend to characterize the biochemical and immunopharmacological activities of the transcript AET-7393 product, classified as a serpin. Several biological processes, such as coagulation and inflammation, are regulated by serine proteases. Thus, our aim is to evaluate the putative targets of this serpin, its role in blood clotting, inflammation and activation of murine macrophages. (AU)
