Abstract

Acute kidney injury(AKI) is a pathology that has high incidence and high morbidity and mortality in the population. Despite all therapeutic advances , these rates are still high. Another alternative, currently suggested would be the transplantation of stem cells. The regenerative process of mesenchymal stem cells (MSCs) has been demonstrated in several experimental models and in some clinical trials. The action mechanism most suggested is the paracrine action of MSCs in the injured area. It is known that in thelesion environment, pro-inflammatory cytokines such as TNFa and IFNg, activate the repair role of MSCs. This study assesses the role of IFNg in the activation of MSC in acute renal models. MSCs of receptor knockout of IFN-g animals (IFN-gR KO) and wild type animals (control / C57/Bl6) will be isolated from adipose tissue. After isolation and its culture, both cells will be administrated in AKI model. In vitro, we will analyze how IFNg activates MSCs, evaluating the products secreted by MSCs, as well as studying signaling pathways and cellular changes resulting from this activation. With this study, in addition to increasing our knowledge about the activation of MSCs, it may create a new therapeutic perspective on which we activated endogenous MSCs, without the need of exougenous MSC administration.
