Abstract

There is considerable interest in establishing therapies that provide a better, faster muscle repair process. A growing body of evidence suggests that low-level laser therapy (LLLT) promotes skeletal muscle regeneration, but its effectiveness in the treatment of injury remains questionable and its therapeutic potential is not fully established. The aim of the present study is to evaluate the effects of LLL on muscle repair and the expression of myogenic regulatory factors (MyoD, Myf5, myogenin and MRF4) following cryoinjury in rat skeletal muscle. Ninety one Wistar rats were divided into five groups: 1) control; 2) sham; 3) cryoinjured without treatment; 4) cryoinjured and LLLT; 5) non-injured and LLLT. The laser irradiation will be performed on the injured region using the GaAlAs laser (780nm) with a beam spot of 0.4 cm2, an output power of 40mW, energy density of 10J/cm2 for 10 seconds. After sacrifice, the Tibialis anterior muscle will be removed for the isolation of total RNA and analysis of myogenic regulatory factors using real-time PCR. Furthermore, muscle samples will be used for immunohistochemical regulatory myogenic factors analysis. The results will be statistically analyzed using ANOVA/Dunnet to determine differences between the control and treated groups. (AU)
