Comparative study of low-intensity lasers and light emitting diode in osteogenesis.

Abstract

Osteoblasts are bone tissue cells capable of producing mineralized matrix. The differentiation of mesenchymal cells into osteoprogenitors is stimulated by several factors and regulated by different signalling pathways and transcriptional regulators. In addition, the MAPK-pathway modulates the cellular proliferation and differentiation. Among the compounds used in the scientific research and bone diseases therapy, the phototherapy with low intensity laser and LED has been investigated with the aim to evaluate its effects on bone metabolism. They, that has specific wavelengths, act in cellular modulation, functioning as a therapeutic agent by adjusting and normalizing cellular activity. Moreover, they have analgesic, anti-inflammatory and promote acceleration of wound healing. Therefore, the aim of this study is to evaluate the comparative influence of phototherapy with low intensity laser (red and infrared lasers) and LED on the proliferation and differentiation of human osteoblasts. Also, we will investigate the role of ERK-MAPK signaling transduction in those responses, by using MEK-specific inhibitor and/or its activation during osteoblast proliferation and differentiation after different phototherapy. Proliferation will be assessed by MTT assay and differentiation by quantifying alkaline phosphatase and the expression of osteogenic/bone formation markers such as Runx-2, osteocalcin and collagen I.
