The effect of using phytotherapics and propolis in physicochemical properties, antimicrobial and biocompatibility of MTA

Abstract

The aim of this study will be evaluate the physicochemical properties, biological and antimicrobial of white MTA mixed with the extracts of Arctium lappa L., Casearia sylvestris SW and propolis. The cements will be divided into six groups, and white MTA mixed with: aqueous extract of Arctium lappa L (G1), 80% distilled water + 20% propylene glycol extract of Arctium lappa L (G2), aqueous extract of Casearia sylvestris Sw (G3), 80% distilled water + 20% propylene glycol extract of Casearia sylvestris Sw (G4), 80% distilled water + 20% propylene glycol extract of propolis (G5) and distilled water (control-G6). To determine the setting time, the standard adopted will be ASTM C266/2008. The flow test will be accomplished according the ISO 6876/2001 specifications. The solubility will be determined volumetrically by microcomputed tomography. In chemical tests to evaluate the pH and calcium release, the materials will be placed in retrograde cavities in acrylic teeth and embedded in and ultrapure water, keeping for 3 hours, 1, 3, 7 and 15 days. This water will be measured for pH and calcium released by the atomic absorption spectrophotometer. To evaluate the antimicrobial effect will be applied direct contact method (Enterococcus faecalis and Candida albicans) to quantifying the colony forming units. Also will be performed the evaluation of intra-tubular decontamination segments of root canals of bovine teeth evaluated by confocal laser scanning microscopy. For the biocompatibility test will be used 162 rats divided into six groups and subdivided into three periods (15, 30 and 60). Each animal will receive two subcutaneous and one alveolar implant (polyethylene tube with cement). After animals death, the samples of subcutaneous and alveolar tissue will be submitted to histologically process to obtain a 5 µm thickness sections and stained with hematoxylin and eosin, the specimens will be proceeded to descriptive and quantitative microscopic analysis. According to the histological results, samples of the alveolar tissue will be processed to analyze the cytokine production by ELISA. Data from each test will be tested for normality by D'Agostino and Pearson, to determine the appropriate statistical test, always with a significance level of 5%. (AU)
