Abstract

This project proposes the chemical, biological and biotechnological studies with two species of Annonaceae (Annona cacans, Annona squamosa, Annona mucosa or Guatteria hispida) in order to search for bioactive compounds in extracts from these plants (fresh and obtained by cell culture). Thus, it will proceed to study the development of cell cultures of these species, and for the most promising, will take a comparative study of plant extracts obtained from fresh and cultured cells.The study of the extracts obtained by both processes will be monitored by the absorption spectrum in the visible region of interest in photodynamic therapy (600 to 800 nm), for assessing the photochemical efficiency, by bioassay and evaluation of the effect as a photosensitizer in photodynamic therapy natural antimicrobial (PDT-antimicrobial). Therefore, it will make adequate methodology and biological testing procedures for antimicrobial PDT (against bacteria and fungi) as well as the development of analytical procedures, including phytochemical study, obtaining the first chemical profile (CCDC, GC, HPLC, NMR or ESI-MS) of bioactive extracts, allowing the identification of some substances known directly in crude extracts. Extracts of two plants in natura most promising step in the screening of substances that can not be identified in step shall be submitted to bioassay-guided fractionation and the substances isolated and purified will perform structural elucidation using spectroscopic methods analysis (IR, UV, NMR and MS spectra). In the biotechnological study will make the development of cell culture of these two plant species in order, in addition to the phytochemical study, the manipulation of physical and chemical factors for the production of secondary metabolites, including bioactive alkaloid liriodenine, carrying out a study comparison of bioactive metabolites in cell cultures and those obtained from fresh plant parts.Thus, it will make the chemical, biological and biotechnological Annonaceae species in order to isolate, characterize and optimize conditions for obtaining extracts and bioactive compounds.
