Abstract

Saccharomyces cerevisiae is widely used to study eukaryotic cell, and is one of the best models for the study of mitochondrial biogenesis. Being a facultative anaerobes, the isolation of mitochondrial mutants is easily obtained by the lack of growth in medium containing non-fermentable carbon sources. Many ORFs of S. cerevisiae have not been characterized yet, including several with possible mitochondrial addressing. We selected seven of this for detailed study: YDR115w and YPR116w, which show clear respiratory phenotype due to instability in the mitochondrial DNA; YDL119c, a probable mitochondrial carrier; YOR022c a possible phospholipase; YPL109c, YLR253w, YLR290c candidates to participate in the biosynthesis of coenzyme Q. To try to elucidate the function of these ORFs, we will study intracellular location and we will search proteic partners through intensification of phenotype or genetic suppressors. The possible formation of protein complexes and association of proteins will also be analyzed by sucrose gradient sedimentation.
