Paper-based platforms for noninvasive detection of tumor biomarkers and cultive neoplastic cells

Abstract

This project aims to create a platform for immunochromatographic assays, which allows a noninvasive detection of tumor biomarkers (sarcosine, BTA, CA 15-3) testing samples of urine and saliva. Along with the immunochromatographic platform, it will be tested the cultivation technique of cells on paper to evaluate the growth profile of neoplastic cells and tumor aggressiveness. The plataform of immunochromatographic assay will receive wax patterns printed by a conventional printer, that creates hydrophobic limits on paper, forming of wax microchannels. For the wax to cross the entire thickness of the paper, there will be a heat treatment, where the increase in temperature will melt the wax, which will go to cross all the paper. After drying, the wax creates a hydrophobic barrier that delimits the microchannel where the reaction occurs. In each microchannel is detected a different biomarker. In each microchannel will be contained specific antibodies immobilized in a region called the test area, another region containing the immobilized biomarker protein in a control zone. Through conjugation of antibodies with colloidal gold nanoparticles will be held immunochromatographic detection of these analytesin the cell culture on paper, it will be used layers of paper with hydrogel containing nutrients to promote cell growth and a plastic mask to separate each microzone. Once created the platform we will add cells from neoplastic tissue and evaluated the potential of this technique to be applied in cell based assays. (AU)
