Abstract

Nowadays, the ER stress has been investigated in cell death process. This cellular compartment can be under stress condition because of protein formation alterations,can participate in ROS production, as well as NADPH oxidase and mitochondria, and promote a reaction cascate from reticulum stress. The aim of this project is to investigate the participation of ER stress in cell death process in neutrophils from diabetic rats. Some parameters will be analysed: the ER in neutrophils by fluorescence microscopy; the ROS production by NADPH oxidade, mitochondria and ER using the amplex red techinique; the caspases 3,4,7 and 12 activities; the content and possible activation or clivages of Bip, IRE1, PERK, ATF-6, ATF-4, eIF2 and ERO1 by western blotting and the expression of the genes Bip, IRE-1, PERK, ATF-6, XBP-1 and CHOP by real time. This study focuses to understand the participation of ER in cell death process of neutrophils from diabetic rats and the possible interation of ER with mitochondria and NADPH oxidase. (AU)
