Senescence is a determining factor in the occurrence of prostate morphological changes, considering the hormonal imbalance and paracrine interactions between epithelium and stroma by means of steroid hormones and different growth factors. However, the role of hormonal therapy and its interactions with the environment prostate is unclear, as well as the dynamics of growth factors in the glandular events in the senescence. Thus, the objective of this study is to characterize and correlate the interactions between different molecules such as FGF-2, FGF-7, FGF-8, EGF, ±-actin and vimentin and AR, ER±, ER² and Prolactin receptors in the epithelial and stromal, separately, in relation to the senescence and hormonal imbalance. Fifty senile male rats (10 month old rats) and ten young male rats (4 month old rats) will be divided into six groups. The Senile /Control group and The Young/Control group will receive 5mL/kg peanut oil every other day for 30 days; The Castrated group: animals will be anesthetized with posterior surgical and chemical castration (10mg/Kg flutamide every other day for 30 days); The Tamoxifen-Letrozol group: animals will receive 1mg/Kg tamoxifen every other day for 30 days and 1mg/Kg letrozol every other day for 30 days; The Castrated + Estrogen group: animals will be surgically and chemically castrated and after will receive 25¼g/kg 17²-estradiol every other day for 30 days; The Tamoxifen-Letrozol + Androgen: animals will receive 1mg/Kg tamoxifen every other day for 30 days and 1mg/Kg letrozol every other day for 30 days and later will receive 5mg/kg testosterone cypionate every other day for 30 days. After the experimental period, the animals will be sacrificed and samples of ventral lobe will be collected. Thirty human prostatic samples will be collected from peripheral prostatic zone obtained from prostatectomized patients (60-90 years old) and divided into two groups: cancer of high grade and cancer of low grade. All samples will be microdissected and submitted to immunological and Western Blotting analysis, including hormonal dosages.
