Abstract

Evaluation of protective mechanisms involved in preconditioning by gentamicin nephrotoxicity in renal proximal tubule cells.The Gentamicin (G) is an antibiotic in the class of aminoglycosides, widely used in the fight against infections. However, she has several side effects and Nephrotoxicity one of the most important. The pathophysiology of acute renal failure (IRA) caused by gentamicin is still unknown, but studies suggest the overproduction of substances vasoconstritoras and decrease of substances vasodilating properties. About 10% of intravenous dose of G accumulates in the renal cortex reaching a much higher concentration of plasma, causing damage to structural and functional changes in Lysosomes, mitochondria and plasma membrane. The preconditioning refers to the situation where a body or cell undergone a ischaemic aggression or toxic acquires resistance or responds less intensely when a new or the same aggression is repeated. It is known that in this facility a number of mediators are involved, after injury, the cell has defense mechanisms cited as "protectors" genes that can express themselves in order to preserve the cell structure and function. Among them are the protein of resistance (MRP-2) and P-glycoprotein that is part of the ATP-binding cassette (ABC) transporters.
