Knowing that the increase of the blood pressure is important in the triggering of an inflammatory response and that the renin-angiotensin system has participation in that, the hypothesis of the present project is that the physical exercise can improve the renal function, acting on the activity of the renin-angiotensin system and, in the crucial form, on the expression of inflammatory cytokines. To respond this hypothesis, will be considered as objective: 1) To evaluate if the time-course of the physical training leads to the morphological renal improvement and if this is connected with the expression of components of the renin-angiotensin system; 2) evaluates the expression of inflammatory markers at the time-course of the physical training. There will be used male rats normotensive (WKY) and spontaneously hypertensive (SHR) originating from the Animal Facility of the ICB/USP, weighting approximately 170-200g in the beginning of the experiment. The animals will be divided in four groups: 1) Sedentary Normotensive (WKY-S); 2) trained Normotensive (WKY-T); 3) sedentary Hypertensive (SHR-S); and 4) trained Hypertensive (SHR-T). Rats were pre-selected for their ability to walk on a treadmill; test of maximum performance were performed before the protocol (week 0), in the 4th one, in the 8th one and in 12th (end) week of training. In the same periods (weeks 0, 4, 8 and 12) rats of the 4 groups will be catheterized in the carotid artery for measure of blood pressure and heart rate. Next, the animals will be killed by overdose of anesthetic and will have the kidneys collected and frozen for dosage of the activity of the angiotensin I converting enzyme (ACE) and of the angiotensins I, II and 1-7, or the kidneys will be perfused for the histological evaluation and detection of renal injury (stained for periodic acid Schiff for evaluation of glomeruloschlerosis and for picrossirius for evaluation of fibrosis), and for immunohistochemical detection of inflammatory markers (interleukines IL-1b and IL-6, and tumoral necrosis factor -TNF-b.). The results will be statistically evaluated comparing the groups for variance analysis (ANOVA) with one or two ways, complemented by appropriately post hoc test. For all the analyses will be adopted the level of signification of p d0,05.
