Abstract

The aim of this project is to investigate hypothetical proteins containing exportation sequence signals and proteins, which show increased expression in the parasite intra-host stages, therefore with potential of being essential for survival of the parasite. The genes will be cloned E.coli in expression vectors, expressed and purified. Antibodies will be generated against them and these used for immunolocalization of the proteins in the parasite. DNA vaccine vectors will also be produced. Mice will be immunized with the recombinant proteins and DNA vaccines and the humoral and cellular response evaluated. The immunized animals will be challenged with S. mansoni cercaria and the reduction in worm burden evaluated by perfusion to establish protection potential. The level and type of immune response will be correlated with the levels of protection obtained to establish the best immune response. (AU)
