Biocátalisys in organic synthesis: Applications of unconventional candidates to biocatalysts in organic reactions

Abstract

The project aims to explore the use of biocatalysts (lipases) as potential promoters of reactions not conventionally promoted by these agents. The proposals are focused on reactions aimed at training liaison carno-carbon (eg Reacões of Diels-Alder), but also links carbon-heteroátomo (eg Michael addition of inter-and intramolecular). It is also exploring alternative synthesis of complex compounds by combining the chemistry of selenium and tellurium as biocatalysis (reaction of the "domino"). The reactions involving reactive unstable entities will be generated in situ under inert and used directly in subsequent steps. The course of these reacões be monitored by thin-layer chromatography and gas chromatography and mass spectrometry (GC-MS). The products of these reactions should be isolated by extractions with organic solvents appropriate and subject to purificações by crystallisation, distillation or by a column chromatography, depending on the characteristics of the substance in question. The reactions biocatalisadas make use of enzymes isolated, plants or microsomes and should be conducted in accordance with the requirements inherent in each case. Similarly to reaçoes not biocatalisadas, monitoring the course of reaçoes should be given the same analytical techniques, but especially by gas chromatography in chiral stationary phase (essential for the verification of the optical enrichment "of products formed). The isolation and purification of the products of these reactions seráo similar to those adopted for the reactions biocatalizadas.Tanto not the products of reactions obtained by conventional synthetic routes such as the reactions biocatalisadas, should be characterized by analytical techniques, which include: Analysis by NMR, 1H, 13C and 15N and possibly 125Te; Analysis of Infra-Red; mass spectrometry (low and high resolution); deviation optical, micro-analysis (CHN), etc..
