This proposal is part of the Researcher Young project approved by the FAPESP that aims at the molecular and functional study of oncoprotein SET, a putative molecular target. This work will be developed in the same Institution and will assist in the establishment of the new area of research that already is being implanted and established in the Faculty of Pharmaceutical Sciences of Ribeirão Preto, USP. The central goal of the project is the investigation of three protein as tumor marker in head and neck cancer. Recently, we validate in tumor cell lines, a new tumoral marker, protein SET (resulted obtained during post doctoral in the NIH; process 05/03380-2). In this proposal we will continue to the study of this tumoral marker in tumors samples from head and neck cancer classified as high aggressive and less aggressive. The same tumors will also be evaluated with regard to the expression of others 2 selected markers (MARK3 and hnRNPK). Important results had been gotten in this first evaluation of the SET, that had shown its participation in one of the more important ways of signalling for tumorigenesis (Leopoldino et al., article in preparation). When we carry through knowdown (RNAi) of this protein in tumoral cell line, the cells had died. This indicated this protein as a therapeutical target potential. This project is part of a contribution with Dr. J. Silvio Gutkind (NIH, U.S.A.) for the continuity of the functional and molecular studies of oncoprotein SET. The development of this work will be very important for the study of the tumors markers chosen aiming at the application of these in the prognostic, diagnosis and accompaniment of the patient, since the recidive happens with great frequency in the head and neck cancer.
