The present study aim to evaluate the microtensile bond strength and the interface morphology of adhesive systems to caries-affected dentin after the use of a papain-based agent. Eighty four human dentin specimens (6 x 6 x 3mm) will be polished, sterilized and randomly distributed into palatal devices of fourteen voluntaries (n=14). Each palatal device will contain 6 dentine slabs and will be used during 14 days according to a design of caries induction by plaque accumulation and sucrose use. After the intra-oral phase, fragments will be removed from the device and randomly assigned into 2 groups according to the caries removal method (I. chemomechanical - Papain-based gel followed by curette) and II. Mechanical - Curette (control group)). Subsequently, they will be divided into 3 subgroups according to the adhesive system tested (Adper Single Bond, Clearfil SE Bond and Clearfil Tris-S Bond). Specimens will be restored with a hybrid composite resin and kept in distilled water at 37 oC, for 24 hours. Then, each resin-tooth block will be sectioned into 5 slabs with 1-mm thickness. One slab of each group will be prepared for the analysis of adhesive interface by scanning electron microscopy (SEM). The remaining slabs will be sectioned again into serial 1-mm2 sticks that will be subjected to tensile stress in universal testing machine (0.5 mm/min). Data obtained from microtensile bond strength will be statistically analyzed by the most adequate method. SEM analysis from the adhesive interface will be qualitatively evaluated. (AU)
