Molecular characterization of cyanobacteria strains isolated from São Paulo State mangrove regions

Abstract

This research project aims to molecular characterize cyanobacterial strains isolated from mangroves regions of the São Paulo State by sequencing their 16S rRNA gene, which has been used to differentiate genetically microorganisms belonging to the Bacteria domain. The sequences obtained will be compared by BLAST (NCBI) analysis and the evolutionary distance between the sequenced nucleotides strains will be analyzed by phylogenetic tree construction. At least fifty cyanobacterial strains will be evaluated. The PCR amplification of sequences of nonribosomal peptide synthetase (NRPS) and polyketide synthase (PKS) enzymes genes will allow to identify among the molecular characterized strains those potentially producing of biochemically active natural compound (cyanopeptides). These enzymes are known to be involved in toxins and antibiotics biosynthesis. The detection of these nonribosomal peptides genes will be carried out using degenerate oligonucleotides primers target to conserved sequences of NRPS and PKS enzymes. The PCR products obtained for these enzymes will be sequenced. These sequences also will be compared by BLAST analysis and their evolutionary relations will be verified by phylogenetic tree construction. The cyanopeptides produced by the strains containing NRPS and PKS genes will be extracted using several solvents. The chemical constituent of cellular extracts will be analyzed using Q-TOF-MS (Quadrupole Time-of-Flight Mass Spectrometry) and HPLC (High Performance Liquid Chromatography). The inhibitory activity of these chemical compounds will be tested against fungus and bacteria.
