Organoid-like intestinal tridimensional (3D) culture for studies on virulence and/or probiotic properties of bacteria and food related compounds

Abstract

Currently, applications of eukaryotic cell cultures are being developed in three-dimensional (3D) systems, which renders organotypic structures that mimics what occurs in vivo. In the case of intestinal cell culture model, it is possible to obtain a highly differentiated three-dimensional structure, which can be very useful for studies of mechanisms of interaction with pathogenic and/or probiotic bacteria, as well as compounds of interest in food.In this project, a model of Caco-2 intestinal cell culture with a Slow Turning Lateral Device, will be started in the Laboratory of Food Microbiology - FCFRP-USP for applications in studies of global gene expression (transcriptomic) to evaluate possible mechanisms of action of pathogenic cultures, probiotics and/or bacteriocins.This proposal is related to a project developed abroad by the applicant with FAPESP support (2016 / 16293-5) at the University of Pennsylvania, USA (January to July 2017) and will promote the dissemination of new knowledge in the institution of origin (FCFRP- USP), as well as strengthen national and international collaborations (AU)
