Abstract

Chemical substances are studied as an alternative or in association with the use of sodium hypochlorite (NaOCl), with the aim of promoting cleaning and disinfection of the root canal system (SCR). Calcium Silicate (CS) is the current perspective for the development of new endodontic sealers. This project aims to study new irrigation solutions and to develop new proposals for endodontic calcium silicate (CS) sealers.Subproject 1- aims to evaluate properties of solutions based on calcium hypochlorite (Ca (OCl) 2) and octenidine hydrochloride (OCT): organic tissue dissolution ability, cytotoxicity, antimicrobial activity and against endotoxins (LPS-lipopolysaccharides) with LPS antigenic potential by using the analysis of inflammatory cytokines (IL-1, IL-6 and TNF-±) from mice.Sub-project 2- aims the development of experimental sealer (ES) based on tricalcium silicate, dicalcium silicate and calcium phosphate, handled with different vehicles. These materials will be subjected to the tests of setting time, pH, radiopacity, solubility and flow, and the sealers that present the best results will be compared to TotalFill BC Sealer (TFBC) and AH Plus (AHP) by using conventional tests and volumetric changes in Micro CT, and the antimicrobial activity. The best experimental sealer (ES) and TFBC will be evaluated with or without association with the substance selected in subproject 1 by using physicochemical, antimicrobial, cytotoxicity and cell bioactivity tests. Biocompatibility and bioactivity in vivo will be assessed by subcutaneous analysis in rats. The CE that shows the best results will be selected.Sub-Project 3- aims to evaluate the flow, filling and penetrability of ES (selected in sub-project 2), TFBC, Neo MTA Plus (NMTAP) compared to AHP cement. The flow will be evaluated according to ISO 6876, and additional evaluation in computerized microtomography (Micro-CT). The filling capacity will be evaluated through Micro-CT analysis. In order to evaluate the penetrability of the cements inside the dentinal tubules, bovine teeth prepared and filled with the cements manipulated with B-rhodium for confocal laser scanning microscope (CLSM) evaluation will be used.
