Evaluation of the antimicrobial effect of alternative and commercial agents on a microcosm biofilm and on prevention of dental demineralization

Abstract

The present study aims to evaluate the antimicrobial and anticaries effect of different commercial agents (natural toothpastes and natural mouthrinses) and alternative (natural extracts and experimental fluoride varnish) in respect to the following parameters: minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), minimum biofilm inhibitory concentration (MBIC), minimum biofilm eradicating concentration (MBEC), microbiological viability (MTT test and/or fluorescence assay using confocal microscopy), production of lactic acid and extracellular and intracellular polysaccharides (EPS/IPS) and/or CFUs counting using a monospecie biofilm (S. mutans) or cariogenic microcosm biofilm formed on enamel and/or dentin, which will be analyzed in terms of mineral loss using transverse microradiography (TMR). All experiments involve individual students' projects, which will take part of this regular grant proposal. In assay 1, commercial toothpastes containing natural agents will be evaluated using microcosm biofilm on enamel in respect microbial viability by fluorescence, production of EPS/IPS and lactate and counting of CFU for cariogenic microorganisms (A.S.B.). In assay 2, commercial mouthrinses containing natural agents will be evaluated using microcosm biofilm on enamel in respect microbial viability by fluorescence, production of EPS/IPS and lactate and counting of CFU for cariogenic microorganisms (A.S.B.). In assay 3, we will evaluate the effect of natural extracts (Myracrodruon urundeuva All and Qualea grandiflora Mart) in respect to MIC, MBC, MBIC and MBEC for S. mutans as well as the viability of S. mutans biofilm formed on enamel by fluorescence (S.S.Z). In assay 4, the same natural extracts will be evaluated using a microcosm biofilm model on enamel, with the analysis of viability using MTT and fluorescence assays, production of EPS/IPS and lactate and counting of CFU for cariogenic microorganisms (J.G.P.). In assay 5, fluoride varnishes (TiF4 and NaF) will be evaluated using microcosm biofilm on enamel and dentin in respect to fluorescence viability, production of EPS/IPS and lactate and counting of CFU for cariogenic microorganisms (D.M.S.S). For all assays, the prevention of demineralization will be quantified using TMR. The data will be submitted to the statistical analysis (p<0.05). (AU)
