Virus inhibitor substances from Nyctaginaceae and Phytolaccaceae native species as plant defense inducers

Abstract

Plant viruses can cause heavy damages and losses in major crops worldwide. Their control is not always efficient. Alternative control measures have been tested, and species containing substances inhibiting viral infection highlighting those from the Caryophyllales order, as Nyctaginaceae and Phytolaccaceae have shown promising results. Brazil has one of the richest flora in the world, and native species have an unexploited potential as antiviral principle sources that could be used. Accordingly, it is intended in this project to evaluate the inhibitory activity of viral infection of native species Guapira opposita (Vell.) Reitz and Pisonia ambigua Heimerl (Nyctaginaceae) and Seguieria langsdorffii Moq. and Gallesia integrifolia (Spreng.) Harms (Phytolaccaceae) as well as to evaluate the effect of plant extracts as plant defense inducers. The leaf extracts from species that present viral infection inhibition superior to 50% in Tobacco mosaic virus (TMV) x Nicotiana glutinosa L. system will be selected for tests in Cucurbita pepo x Zuchinni yellow mosaic virus (ZYMV), which presents systemic infection. The extracts will be evaluated at different dilutions as well as at time intervals between treatment and ZYMV inoculation. Other aspects that will be evaluated refer to the parameters related to defense signaling in C. pepo, such as hydrogen peroxide, nitric oxide, salicylic and jasmonic acids and polyamines in four treatments: (i) plants treated with leaf extract(s) from Nyctaginaceae and Phytolaccaceae selected species, (ii) plants treated with phosphate buffer only, (iii) plants inoculated with ZYMV and (iv) treated and inoculated plants. In addition, the metabolomic profile as well as expression of the gene coding for proteins related to the pathogenesis (PR) of C. pepowill also be evaluated in the four treatments by means of gas chromatography coupled to mass spectrometry and qRT-PCR, respectively. The realization of this project will not only open new perspectives of alternative control of economically important plant viruses through the use of native species of the Brazilian flora but will also provide information for studies related to the mechanism of action, as well as the interactions in the defense processes as reactive oxygen species elicitors, PR gene expression and cascade of defense-related reactions.
