Study of sexual chromosomes markers in the São Paulo state population: 32 X-InDels and 23 Y- STRs analysis

Abstract

The autosomal STR markers analysis is considered the standard method used in human identification and forensic genetics, therefore, has a high power of discrimination which is enough to solution the majority of cases. However, in specific cases, such as resolution of a paternity case where the alleged father is absent and genetic reconstruction by close family members is required, only the autosomal STRs analysis may not be enough to obtain a conclusive result. So it is necessary the analysis of sex chromosomes X and Y markers to complement the results of autosomal markers and get a conclusive report. Furthermore, is common in this context the analysis of DNA samples degraded and/or contaminated. In these cases, there may be analytical difficulties of STR markers and recommends the use of different types of markers, for example the InDels, which have a smaller fragment to be analyzed in PCR what increases the chance of success in the analysis of such material. In this project, will use two sets of sex chromosomes markers ( 32 X-InDels and 23 Y- STRs) to study the population of São Paulo for the testing of different forensic parameters. The genetic diversity existing in individuals of the same group and between different population groups, can be evaluated from the distribution of allelic frequencies of molecular markers. It should be emphasized that this type of research can not only be considered as merely technical, because the frequencies of information is extremely important in solving criminal cases and/or complex paternity, which in both situations involve feelings, loss, inheritances and human dignity. Therefore, the project is very important because it allows the transfer of technologies to the community , one of the main goals of academic scientific research. Therefore , these data will more statistical support for the use of these methodologies in forensic identification and / or paternity identification for the population. (AU)
