Analysis of the carbonic anhydrase VI and ±-amylase activity in saliva and biofilm after a cariogenic challenge with sucrose and starch in children aged 3 to 5 years

Abstract

Objectives: To study the activity of carbonic anhydrase VI (CA VI) and ±-amylase in saliva and in biofilm after a cariogenic challenge with sucrose and starch in children aged 3 to 5 years. Methods: Fifty-four (54) preschoolers aged 3 to 5 years, both genders, will be divided into 2 groups: caries free (CF, n = 27) and caries (C, n = 27). In each individual, the presence of visible biofilm in the upper incisors will be verified as well as the caries index, the stimulated saliva flow rate, the buffer capacity and frequency of sugar intake. The volunteers will be exposed to rinses with 20% sucrose, 2% starch, and 20% sucrose + 2% starch solutions. In Chapter 1, the behavior of AC VI and ±-amylase activity will be analyzed in saliva before and after the cariogenic challenges. In Chapter 2, the enzymes activity will be investigated in biofilm samples, as well as pH. In Chapter 3, children will be instructed to perform the three types of rinses, under different experimental conditions. Individuals will be subjected to professional prophylaxis for the biofilm removal + 2 days of regular brushing (2x / day) (phase 1), and at subsequent times, children will be asked to refrain from brushing their teeth for 48 hours (phase 2). AC VI activity analysis will be performed in saliva and biofilm using zymography. ±-amylase activity analysis will be performed by enzyme-linked immunosorbent assay (ELISA). Salivary flow rate will be expressed as mL/min. The pH of saliva and biofilm will be measured with a specific electrode. The saliva buffer capacity will be measured using microelectrode. Statistical analysis: After normality test, non-parametric data will be transformed and submitted to the ANOVA test, followed by the Bonferroni test. Pearson correlation analysis (r), and Multiple Linear Regression (r2) analysis will be performed between variables. (AU)
