Vesicle trafficking during heparan sulfate biosynthesis

Abstract

Heparam sulfate (HS), a sulfated linear carbohydrate that decorates the cell surface and the extracellular matrix, is widely distributed among the various phyla of the Animal Kingdom and represents a key regulator of biological processes. Its biosynthesis, in the form of proteoglycans (PGs), occurs mainly in the lumen of the Golgi apparatus, after its initiation in the lumen of the endoplasmic reticulum (ER) where the first monosaccharide is added to a serine residue. The action of several enzymes along the Golgi generates distinct substitutions patterns and, ultimately, a tremendous structural diversity. Yet, while conserved from yeast to higher animals and plants, the vesicle trafficking between the ER and Golgi as well as its regulation during HS biosynthesis remains hitherto unknown. Thus, by coupling structural analyses to cell biology tools, we will investigate the possible biosynthetic routes involved in HS biosynthesis (AU)
