Adsorption of bisphosphonates and proteins on TiO2 surfaces for bone implants

Abstract

Nanostructured titanium dioxide (TiO2) has been used as surface modifier in biomedical implants, promoting biocorrosion resistance and increasing bioactivity of the oxides, showing promising results with the interaction with living matter. Although the biocompatibility of this oxide is recognized, there are still many aspects of the adhesion mechanisms between biomolecules and the surface of the material there are some open questions that must need to be better understood. Chemically the surface of these oxides is mostly terminated by OH groups, which can be readily functionalized with bifunctional organic molecules. This chemical functionalization or simply a physical modification of the surface can improve the interaction of the oxide with the biological environment, improving bone resorption in orthopedic and dental implants.This project proposes to synthesize TiO2 thin films on metallic titanium substrates and to study different functionalization strategies of surfaces using biofunctional molecules such as bisphosphonates and proteins. It is also intended to evaluate the physico-chemical and thermodynamic parameters in order to analyze the better interaction and adsorption between protein / bisphosphonate and TiO2 surface, since different biomedical applications require different protein-oxide interaction properties. Different strategies of functionalization and adsorption will be studied and tested in biological systems in vitro. (AU)
