Evaluation of fungal infections and characterization of yeast in liver transplanted patients

Abstract

Infections are the most frequent and serious complications after liver transplantation, among them are those caused by fungi, being more serious when invasive. Objectives: to evaluate the incidence of fungal infections, to characterize yeasts of the genus Candida present in the oral cavity and to perform an antifungal sensitivity test on the identified yeasts. Method: A cross-sectional study will be carried out between May 2017 and May 2018, at Hospital São Vicente de Paula, Passo Fundo, Rio Grande do Sul. Patients (n = 96) older than 18 years who agree to participate in the study will be included and will be assisted by the Intra-Hospital Organ Donation and Transplant Tissue Commission (CIHDOTT). Sociodemographic and clinical data will be collected and during the physical examination performed by the medical staff of the transplant sector. Material will be collected from patients who present lesions suggestive of fungal infections and oral material from all patients once through the swab technique and this will be seeded in Petry dishes containing Sabouraud Agar culture medium plus chloramphenicol. All fungi will be identified for morphological and molecular aspects. Phenotypically, filamentous fungi will be identified according to classical parameters of international characterization of fungi and yeasts through CHROMagar ® chromogenic culture medium (DIFCO), and by means of method ID 32 - C - Système d'identification des levures - BioMérieux AS. Molecular identification of fungal species will be performed by PCR amplification and DNA sequencing. After the antifungal sensitivity test for all fungi will be performed through the broth microdilution methodology, according to the standards of the Clinical and Laboratory Standards Institute (NCCLS) M27-A3.
