Flavonoids combinations in thermosensitive hidrogels as proposals of endodontic medications for young permanent teeth


The objective of this study was to evaluate the in vitro antimicrobial, antiinflammatory and inductive effects of mineralization of compounds representing the main classes of natural, isolated and combined flavonoids and to study proposals to incorporate these compounds into thermosensitive hydrogels as endodontic medication. The aim of this study was to evaluate the antimicrobial activity of the following flavonoids: taxifolin, pinocembrin, galangin, chrysin, EGCG, quercetin, kaempferol, myricetin, naringin, genistein, daidzein and silibilin, by determining MIC, CBM and CIF on bacteria of endodontic interest. Then, flavonoids with antimicrobial activity, either alone or in combination, will be selected to evaluate the effect on bacterial biofilm by counting the CFU / mL. In macrophage culture, the flavonoids will be screened for cell viability (MTT assay) and subjected to ROS quantification, NO inhibition and qPCR assays for determination of iNOS, COX-2 and IL-1² expression levels. In addition, the compounds will also be evaluated on undifferentiated pulp cells and osteoblasts for the induction of mineralization by determination of ALP activity, quantification of nodules of mineralization and expression of DMP-1, DSPP, OPG, RANKL and OCN. The flavonoids or combinations of compounds with the best results in the above assays will be incorporated into two types of thermosensitive hydrogels: PNVCL or chitosan and evaluated the antibiofilm effect in confocal microscopy and cytotoxicity by means of MTT test and fluorescence microscopy. This study intends to propose an intracanal medication with natural flavonoids with therapeutic properties based on the in vitro results and that may have future clinical application. (AU)
