Antimicrobial photodynamic therapy (aPDT) in the treatment of periodontal disease induced in rats

Abstract

Therapeutic alternatives have been developed to aid treating of periodontal disease (PD), and among them Antimicrobial Photodynamic Therapy (aPDT) highlights. This study aims at evaluating the effect of aPDT on the bacteria present in the context of periodontal disease as well as its effects on the dental supporting tissues. The aPDT will be carried out using FS hypericin-glucamine and Phthalocyanine-glucamine activated by LED in the treatment of PD induced in rats. In "Study 1", the microorganisms involved in the ligature-induced PD and influence of aPDT will be assessed by microbiological analysis. The "Study 2" will evaluate the supporting tissues by the following techniques: microtomography, histometry, stereometry, histology and immunohistochemistry. Ligatures of cotton yarn will be inserted in the grooves of the upper second molars of both maxillary for induction of the PD, remaining in the position throughout the experimental period. The animals will be randomized into 6 groups and the following treatments will be held 7 days after inserting the ligatures: G1. Hypericin-glucamine; G2 - Phthalocyanine-glucamine; G3 - Amber LED; G4 - Red LED; G5 - aPDT with Hypericin-glucamine; G6 - aPDT with Phthalocyanine-glucamine. The hemimaxila who receive the treatment will be chosen randomly and the opposite hemimaxila the negative control (no treatment) of each animal. Groups belonging to the period of 7 days will be euthanized with anesthetic overdose on the same day of treatment. At 7, 15 and 30 days after the induction of periodontal disease, the animals will be euthanized."Study 3": The objective of this study is to evaluate in vitro antibacterial effect of APDT using hypericin-glucamine photosensitizer and phthalocyanine-glucamine culture of anaerobic Gram negative.The results will be analyzed statistically.
