Real-time investigation of heat produced in the metabolic activity of erythrocytes of patients with sickle cell anemia in normoxia and anoxia conditions, using a segmented flow chip-calorimeter


The request for this visiting researcher project - is the target of a joint project of the groups of Prof. Pedro L.O. Volpe (Institute of Chemistry - Unicamp) and the research group of Prof. Dr. Fernando Ferreira Costa (Hemocentro - FCM - Unicamp), Dr. Carolina Lanaro, Dr. Dulcinéia Martins de Albuquerque, and Prof. Dr. Kleber Y. Fertrin (Department of Clinical Pathology) with the group of Prof. Dr. Johannes Lerchner (Technische Universität Bergakademie - Freiberg - Germany) research for the development of new calorimetric methods and their application for the direct determination of metabolic activities in blood cells in real time. We have previously found (FAPESP 14/1781) that erythrocytes from sickle cell patients (SS-RBC) produce about twice as much heat in comparison to red blood cells from healthy patients (RBC). However, it was not clear to what extent the actual sickling process contributes to increased heat production. Therefore, a new microcalorimetric investigation on sickle cell SS-RBC is proposed to compare the rate of heat production, that is, the metabolic activity of the erythrocyte sample in the falciform and non-falciform state. Therefore, for the first time, changes in the rate of heat production dependent on the decrease or increase in the supply of oxygen can be analyzed in real time since a new Chip calorimeter built in the research group of Professor Lerchner allows the establishment of well defined tensions of oxygen in blood cell samples. To do this, a new version of the segmented flow Chip microcalorimeter, built to submit the segments of blood samples under conditions of normoxia and anoxia, will be installed at the Institute of Chemistry of UNICAMP during the duration of the project. The focus of the project is focused on the use of microcalorimetry in the study of the variation of the red cell metabolic activity of patients with sickle cell anemia as a function of oxygen tension using the Chip Microcalorimeter in the Segmented Flow Technology.
