TOBACCO SMOKING: FROM MOLECULAR ASPECTS TO SMOKER APPROACHES

Abstract

This aim of this work is to investigate smoking as an addiction till different molecular changes caused by tobacco. Molecular and cytogenetic analysis will evaluate the tobacco use damage. Four groups will be investigated for molecular and cytogenetic changes; Group 1: 30 patients with oral squamous cell carcinoma (OSCC); Group 2: 30 patients equivalent to group 1 with benign lesions in oral cavity, group 3: 30 electronic cigarette smoking patients, Group 4: 30 smokers of manufactured cigarette. Gene expression will be held in groups 1 and 2 by qPCR for metabolizing genes of tobacco carcinogens: CYP1A1, CYP1B1, CYP2E1, CYP2A6, GSTM1, GSTP1 e GSTT1 and to the repair genes MLH1, MSH2, ATM, MRE11A, PMS2, XRCC1 and MLH3. Both the CYP family genes and GSTs are part of the xenobiotic metabolism and excretion system. For these groups, PCR Array will also be performed for assessing methylation in 24 tumor suppressor genes. The PTS Detect " Cotinine System® (PTS Diagnostics, Indianapolis, USA) will be used to evaluate nicotine concentration in the capillary blood of patients in group 3, for an objective test of consumption.All cases results of OSCC will be correlated with the clinical and histological data. The genotoxic effect of the electronic cigarette will be evaluated on groups 3 and 4, by the micronuclei research in exfoliated 3000 buccal mucosa cells and peripheral blood lymphocytes. The statistical analysis of all proposed methods of evaluation, it will be realized by parametric Student t test. For nonparametric data Mann-Whitney test will be used. The association between the variables will be analyzed by chi-square test and the correlation with tobacco consumption by Pearson correlation. (AU)
