Karyotype and Sex Chromosomes Evolution in Neotropical Fishes. Part II.

Abstract

Fishes from the Erythrinidae and Triportheidae families are excellent investigative models for evolutionary cytogenetics. An outstanding collection of karyotypic data and chromosomal markers, particularly from genus Hoplias (Erythrinidae) and Triportheus (Triportheidae) have been obtained in our previous studies. This project will continue the previous one (FAPESP- 2014 / 22532-7), now focusing on the interrelationships of these families with other fish groups in order to achieve further progress with regards to the karyotype and sex chromosomes evolution. Regarding the Erythrinidae family, their chromosomal relationships with the Hepsetidae, Lebiasinidae and Ctenolucidae will be investigated, considering the phylogenetic proposals that points out their relatedness. We will use resolute chromosomal markers through molecular cytogenetic analysis, aiming to clarify the evolutionary relationships between these families, a still unexplored field in Neotropical Ichthyology. There will be also emphasized the genesis and differentiation of a rare XX/XY1Y2 system found among fishes, thus advancing on the understanding of the evolutionary process of the sex chromosomes among the lower vertebrates. In turn, the prospect of homologies and genomic differences between the karyomorphs of the H. malabaricus species complex will provide a significant step in the characterization of a cryptic biodiversity present in our fish populations. To this end, comparative genomic hybridization (CGH) and chromosome painting (WCP) with probes derived from chromosome microdissection procedures will be applied. Regarding the Triportheidae family, comparative cytogenetics of Triportheus species with other congeneric groups, as well as sister families, will investigate the evolutionary genesis of the ZZ/ZW system chromosome, characteristic of Triportheus, among related groups. Significantly, the presence of the same sex system in closely related groups would highlight the trajectory of sex chromosomes among fishes, revealing an evolutionary novelty in this biological group. For this purpose, CGH and WCP experiments with probes derived from sex Z and W chromosomes of Triportheus, will be applied. Finally, the search for genes associated with Triportheus sex chromosomes will provide significant subsidies for future establishment of sex determination candidate genes, a field still highly lacking in knowledge in this biological group. In this sense, next generation sequencing thought the DART-Seq method, highly informative and with unprecedented application in fishes, will be applied in partnership with researchers from the University of Canberra (Australia). Therefore, the expected results of this project should enable significant advances regarding the karyotype and sex chromosomes evolution, characterization of cryptic biodiversity of our fish populations, their evolutionary relationships and their genomic composition, taking advantage of previous knowledge acquired with the two models groups that support this proposal.
