Effect of a protocol of decontamination associated with L-PRF in induced periimplantar defects in swine model.

Abstract

To date, there is no gold standard treatment for periimplantitis. Then, the establishment of an effective therapeutic protocol for decontamination and re-osseointegration is needed. The aim of this study is to characterize the microbiological, inflammatory, clinical and radiographic profile of induced periimplantitis in minipigs, and evaluate the effect of antimicrobial photodynamic therapy (aPDT) associated with L-PRF in decontamination and re-osseointegration of periimplant defects. The mandibular premolars of 8 minipigs will be extracted bilaterally, preceding the insertion of 8 dental implants. After a healing period, ligatures will be placed for 2 months for induction of the experimental periimplantitis and 15 days before treatment, ligatures will be removed. Two antiinfective protocols will be used to decontaminate implant surface: mechanical treatment associated or not to L-PRF, aPDTassociated or not with L-PRF. Twelve weeks post treatment, the implants will receive healing abutment. To characterize the periimplantitis will be performed the analysis of cytokines (inflammatory profile), and pyrosequencing (microbiological profile), radiographic assessment of bone loss and clinical progression of the disease. Treatment effect will be evaluated by means of clinical data, analysis of the defect extension, micro-CT, histometry and histomorphometry analyses. It is expected that the implants treated with aPDT and L-PRF presents higher decontamination indexes and better re-osseointegration rates. (AU)
