Molecular techniques for validation of reproductive biotechnologies in fish used for preservation of endangered species

Abstract

The agricultural and industrial activities, as well as the urban development, results in increased riparian area deforestation, silting of rivers and changes in their courses and increased emission of pollutants in the waters of the continental rivers. Freshwater fishes are then subject to all these environmental impacts, which may lead to the extinction of many species. Some reproductive biotechnologies, such as androgenesis, gynogenesis, cloning and chimerism, are applicable to recover the natural fish stocks and prevent their extinction. Most of these techniques are currently being applied in the Laboratory of Fish Biotechnology at National Center Research for Conservation of Freshwater Fishes (CEPTA-ICMBio) in Pirassununga - SP, including master, doctoral and postdoctoral members associated to the UNESP and USP universities. All the mentioned reproductive biotechnologies must be evaluated by molecular tools in order to confirm the effectiveness of those techniques, such as paternity testing, in vitro transcription, cloning and gene sequencing and polymerase chain reaction using species-specific primers. The Laboratory of Fish Biotechnology at CEPTA-ICMBio presents part of the equipment required to develop the molecular analysis. Therefore, this project aims seeking out for funding to assess adequately the laboratory for molecular technologies and that includes the acquisition of kits, reagents, enzymes and materials for molecular analysis. CEPTA-ICMBio is an important Brazilian institution for the conservation of continental aquatic biodiversity and it has collaborated in many projects with various universities. The establishment of the Molecular Biology Laboratory, associated with the Laboratory of Fish Biotechnology will assess the reproductive techniques and it will provide increased impact of researches, as well as the conclusion of dissertations and theses in this institution.
