Abstract

The creation of horses has been actively developed in Brazil, so the owners' interest in equine reproductive biotechnologies is increasing, such as artificial insemination (AI) with fresh, cooled and frozen semen. However, the equine semen during handling lose quality and fertility, and oxidative stress an important cause of damage to the sperm cell. Seminal plasma has protective effects on the membrane of the sperm and prevent premature capacitation of them during the cooling process and cryopreservation. Since the removal of the seminal plasma during these processes increases oxidative stress. Antioxidants without seminal plasma promote an important role in sperm protection. Carnosine is one of those substances that acts on the reduction of the shortening rate of telomeres. The purpose of this study and identify and quantify carnosine in seminal plasma and compare how the fresh semen characteristics, cooled and thawed between the stallions will be classified according to seminal features fresh good and poor quality. Forty stallions will be collected twice (aged 4 to 15 years of age) in November 2016. The fresh semen, cooled and thawed will be evaluated for motility, vigor, concentration, acrosome integrity and plasma membrane, mitochondrial activity (SCSA ) for susceptibility to denaturation acid chromatin and TBARS (thiobarbituric acid reactive substances). Carnosine of seminal plasma will be measured using a commercial ELISA kit. (AU)
